Sequence Automation In This Section, We Will Discuss: How to set up a sequence. How to perform automated recalibrations. How to do bracketing. How data files are named. How to use Sequence Summary. How to run the sequence. The sequence container (folder). 2 Building a Sequence for Automated Analysis 1. Go to the Method and Run Control view. 2. Go to View > Preferences and set up sequence folder naming convention. 3. From the Sequence menu, select New Sequence. 3. Edit the Sequence Parameters. 4. Edit the Sequence Table. 5. Select a Sequence Output. 6. Save the Sequence 7. Run the Sequence form the RunControl menu or the Sequence Table. Note: Hypersequence is a feature for the Agilent 1100/1200 Series Well plate Handler only. 3 Preferences •Specifies naming convention for sequence data containers. 4 Sequence Parameters 5 Explicit Calibration Calib Calib Calib Samples Samples Standards Standards Standards Update Calibration Update Calibration Calculate Sample Amounts After Each Injection Update Calibration Calculate Sample Amounts After Each Injection 6 Sequence Table Explicit Recalibration Additional Columns in Sequence Tables: •Sample Amount •ISTD Amount •Multiplier •Dilution •Data file •Inj Volume •Lims ID •AutoBalance 7 Customize Sequence Table 8 Explicit Plus Cyclic Recalibration 9 Bracketing Calibration Injection Calib Calib Calib Samples Samples Standards Standards Standards Update Calibration Update Calibration Calculate Sample Amounts For Each Injection Calculate Sample Amounts For Each Injection 10 Bracketed Sequence Operation 11 Partial Sequence Preview of Bracketing Will show you the order of injections. 12 Fill in Sequence Table Using Sequence Table Fill Down Wizard 13 Sequence Output A Sequence Summary Report allows you to select from a simple one page Sample Summary to a comprehensive report documenting all aspects of the analysis including crosssample statistics. 14 Edit Sequence Summary Statistics calculated in Standard or Extended format. 15 Extended Statistics 16 Save the Sequence File Save Current Sequence 17 Sequence Sampling Diagram Shows sequence tray set-up and sequence processing status. Click to set-up sequence. Select Start to begin sequence. Select from the View menu. 18 Start a Sequence •RunControl menu •Start button •F6 •Sequence Table •Partial Sequence Table •When you start a sequence, a sequence folder is created in the DATA directory. •The sequence folder is named according to the subdirectory specified in Sequence Parameters, and the naming convention specified in Preferences. •A copy of the master methods (Chem32\1\Methods\names.M) and sequence template file (Chem32\1\Sequences\name.S) are copied to the folder. The sequence acquisition is run from these copies. 19 Sequence Folder Data Structure Sequence Container (Folder) Subdirectory sequencetemplatename year-month-day hour-minute-second C:\Chem32\1\DATA\03-15-06\CLASSSEQUENCE 2006-03-15 10-46-09 Acquired Data Copy of Master Method Batch file for Batch Review Copy of Sequence Template Sequence Logbook 20 Sequence Data Files Format for Automatic Injection: 003-0101 003 Vial Number 01 Sequence Line 01 Replicate Prefix Prefix000001 Prefix000002, etc. for a total of 15 characters Format for a Cyclic Calibration Standard: C1-02001 C Indicates a cyclic calibration standard was used 1 Calib Line number 02 Sequence Line 001 Replicate 21 Recalibration Settings 22 Calibration Report Options 23
