10-1 Squalamine Family of Aminosterol Antibiotics from Various Shark Species The FASEB Journal. 2007;21:791.4 Squalamine is a sulfated aminosterol, originally detected in Squalus acanthias (spiny dogfish shark) due to its broad-spectrum antimicrobial (antibiotic) activity. Large-scale natural product extraction of spiny dogfish liver has established that squalamine can be isolated in considerable quantities, along with smaller amounts of structurally related aminosterols. In addition to antibiotic activity, each aminosterol has another remarkable pharmacological property, such as antiangiogenesis activity, appetite suppression, or inhibition of sodium proton exchanger proteins. These water-soluble steroids have therapeutic potential in treating various human diseases. To investigate the biosynthesis of these compounds, a mini-scale extraction scheme was devised first to determine their subcellular localization. In spiny dogfish liver, both the total membrane and grey fat fractions are very rich in these amphipathic aminosterols.
