Pgp knockout mice also are used for verification of the in vitro conclusions.

A survey of successful CNS drugs indicated
ER < 1 (22%), ER 1–3 (72%), ER > 3 (6%)
This suggests that many commercial CNS drugs are subject to low-to-moderate
levels of Pgp efflux that can be overcome with clinical dosing.
However, high Pgp efflux (ER > 3) is uncommon in commercial CNS drugs and
should be avoided.
